Can i connect the cable directly to my notebook Ethernet port from Desktop computer (Ethernet port) to transfer ( 20 Gb file )

No. You cannot do that. Just by wiring two computers you cannot transfer a file. For two computers to talk to each other over a network both require an independent IP address. Only a router can provide unique IP address to each computer in its network.

So if you have a wired or wireless network at home and both are connected to it you can do it by folder sharing.
